Ipamorelin is a short synthetic peptide that contains several non-proteinogenic building blocks. Precisely these building blocks determine which analytical methods can sensibly be applied.
What is Ipamorelin?
Ipamorelin is a pentapeptide with the structure Aib-His-D-2-Nal-D-Phe-Lys-NH2 and a molecular mass of approximately 712 Da. It contains three particularities: the non-proteinogenic amino acid aminoisobutyric acid at the N-terminus, two amino acids in D configuration including the unnatural 2-naphthylalanine, as well as an amidated C-terminal end.
The C-terminal amidation reduces the mass compared with the free acid by approximately 1 Da. This small difference can be resolved by mass spectrometry and serves as a check on whether the amidation is actually present.
Research context
Ipamorelin is described in the literature on growth hormone secretagogues and is frequently investigated there together with GHRH analogues such as CJC-1295. An approved medicinal product does not exist. For analytical questions it is relevant that classical methods such as amino acid analysis or Edman degradation function only to a limited extent with this structure, because they are designed for proteinogenic L-amino acids and a free N-terminus.
Analytical identification
- RP-HPLC on C18 phases is well suited. The naphthyl residue provides pronounced hydrophobicity and therefore clear retention. With naphthylalanine and phenylalanine, ipamorelin also possesses aromatic systems, so that in addition to detection at 214 nm, detection in the higher UV range is possible.
- LC-MS confirms the molecular mass of around 712 Da, usually as a singly charged ion. Fragmentation in tandem MS additionally allows the sequence order to be secured.
The D-configured amino acids cannot be distinguished from their L-enantiomers by mass spectrometry, since they have the same mass. A mix-up of the configuration would remain undetected in a pure LC-MS run and could only be demonstrated via chiral analysis or via the retention time against a reference standard. This is a good example of the fact that even the combination of HPLC and MS does not cover every question.
Purity and analytical interpretation
With pentapeptides, deletion sequences are usually well separated chromatographically from the main peak, which makes interpretation easier than with long peptides. Typical by-products are incompletely amidated species as well as epimerisation products. A stated area-percent value remains here too a statement about the chromatogram, not about the content of the sample; residual salts and residual solvents are not recorded in the UV.
Documentation and batch traceability
Traceable documentation names the method, detection wavelength, measured mass and batch reference. With peptides containing unnatural building blocks it is additionally helpful if the structure is stated in full, since the trade designation does not document the configuration.
Storage and Stability
Lyophilised material is stored dry, protected from light and refrigerated, for longer stocks usually at minus 20 degrees Celsius. Freeze-thaw cycles are to be avoided. These details concern exclusively the handling of laboratory substances.
