There is no aesthetic motive behind this, but a process: lyophilisation, colloquially freeze-drying.

This article explains the physical principle, the three phases of the process, why the method is chosen for sensitive molecules, and what the appearance of a lyophilisate can indicate about the course of drying.

The principle: sublimation instead of evaporation

In ordinary drying, water passes from the liquid to the gaseous state. In lyophilisation this intermediate step is bypassed: the water is first frozen and then passes directly from the solid to the gaseous state under reduced pressure. This process is called sublimation.

The decisive physical reference point is the triple point of water. Below the associated pressure there is no longer a liquid phase; ice can then only sublime. This is precisely the range in which a freeze-dryer operates. The advantage: the material does not pass through a liquid phase at elevated temperature, which considerably reduces mechanical and thermal stress.

The three phases of the process

Phase

What happens

Critical parameter

Freezing

The solution is cooled until it solidifies completely; ice crystals and a concentrated residual phase form

Cooling rate and final temperature; they determine the crystal size and thus the later pore structure

Primary drying

Under vacuum the frozen water sublimes; most of the moisture is removed here

The product temperature must remain below the critical temperature of the material

Secondary drying

The residual water bound to the matrix is removed by desorption

Temperature and duration; determines the final moisture content of the lyophilisate

The phases build on one another. If primary drying is run too warm, the frozen structure can partially collapse; the product collapses and no longer forms a stable cake. If secondary drying is shortened, more residual moisture remains. Both effects are partly visible on the final product.

Why freeze-drying is chosen for peptides

Peptides are markedly more reactive in aqueous solution than in the dry state. Water is both a reaction partner and a transport medium: hydrolysis, deamidation and oxidative processes proceed considerably faster in solution. Removing the water slows these reaction pathways down considerably.

Reduced molecular mobility: in a dry, amorphous matrix molecules are strongly restricted, and chemical reactions correspondingly slowed.

No thermal stress: the entire process runs at low temperatures, in contrast to heat-drying methods.

Defined mass per container: the solution is filled before drying, so that the solid forms inside the container.

Porous structure: the resulting cake has a large internal surface area and can be redissolved comparatively well.

Which chemical changes are meant specifically - such as oxidation at methionine or deamidation at asparagine - is described in the article on Impurities in Synthetic Peptides page.

What the appearance of a lyophilisate suggests

The appearance is a rough but not worthless first indication of how drying proceeded. It does not replace analytics and allows no statement about purity or identity.

Observation

Possible meaning

What it does not mean

Loose, dimensionally stable cake

Drying carried out below the critical temperature

No proof of a particular purity

Collapsed or glassy structure

Possible collapse during primary drying

Not automatically an indication of degradation

Very small, barely visible quantity

Normal for small weighed amounts, in particular below 1 mg

No indication of a missing fill

Material distributed on the rim of the container

Movement during transport or filling

Not a quality defect in itself

The third row is practically relevant: with very small quantities of substance the visible solid is often barely perceptible. The peptide quantity actually contained does not follow from the visual impression, but from the weighed amount and the net peptide content; on this see the article on net peptide content.

Lyophilisate and stability: the connection

Freeze-drying creates a state that slows chemical reactions down - it does not make a material keep indefinitely. Two factors remain decisive: the residual moisture left behind and the conditions under which the sealed container is subsequently kept. A hygroscopic lyophilisate takes up moisture from the surrounding air as soon as the container is opened.

The relevant environmental parameters are covered in the article on storage and stability in the laboratory; the conditions during transport are described in the article on cold-chain shipping.

Frequently Asked Questions

Is a collapsed cake grounds for complaint?

A collapse points to a drying process above the critical temperature and can be accompanied by higher residual moisture. Whether this is relevant for a specific batch can only be clarified via the associated batch documentation, not via the visual impression.

Why are peptides not simply supplied in solution?

Because reactions such as hydrolysis, deamidation and oxidation proceed considerably faster in an aqueous environment. The dry state is the more stable state for storage and transport.

Does a lyophilisate still contain water?

As a rule yes, to a small extent. Complete dryness is practically unattainable. The remaining residual moisture is, where tested, determined by methods such as Karl Fischer titration and enters into the content calculation.

What does the term “cake” mean in this context?

The porous solid structure that remains in the container after sublimation is referred to as a cake. Its shape arises as an image of the original ice-crystal structure.

Does freeze-drying say anything about purity?

No. Lyophilisation is a drying process, not a purification process. Statements on purity come from chromatography, statements on identity from mass spectrometry. The underlying system is set out in the article on Purity, identity and content .

Sources and further reading

European Pharmacopoeia (Ph.
