The attached fatty acid chain changes the chromatographic behaviour considerably and imposes its own requirements on the analysis.

What is semaglutide?

Semaglutide is a peptide of 31 amino acids derived from human GLP-1. Compared with the natural sequence two positions are exchanged: at position 8 stands the non-proteinogenic aminoisobutyric acid, at position 34 arginine. A C18 diacid is bound to the lysine at position 26 via two AEEA spacers. The average molecular mass is approximately 4,114 Da.

The combination of peptide backbone and long-chain fatty acid makes the molecule amphiphilic. Chemically it is therefore not a simple peptide but a lipopeptide conjugate.

Research context

Semaglutide is extensively described in the scientific literature as an incretin mimetic. In this catalogue corresponding compounds are listed exclusively as reference and research material; they are not intended for use in humans or animals and this page makes no statements about effect, indication or application. Analytically it is relevant that peptide analogues of this class are structurally closely related and in part differ only by a few amino acids or by the length of the fatty acid chain.

Analytical identification

RP-HPLC requires a considerably higher organic proportion than with unmodified peptides, since the C18 diacid interacts strongly with the stationary phase. Without an adapted gradient the substance appears late, broad or not at all. The method parameters are therefore not a detail here but a precondition for an interpretable chromatogram.

LC-MS provides the mass confirmation. At approximately 4.1 kDa a multiply charged ion series arises in electrospray; the mass is calculated by deconvolution. The mass difference from related analogues is large enough to exclude a mix-up, provided a mass measurement is available at all.

Since structurally similar analogues are traded in parallel on the market, an identity check via the mass is more informative than a purity figure alone.

Purity and analytical interpretation

Typical related substances are incompletely acylated species, conjugates with the wrong number of spacers as well as deamidation and oxidation products. Some of these by-products differ only slightly from the main peak chromatographically. A single area-percent value without a chromatogram, gradient and wavelength is therefore not verifiable and says nothing about the peptide content of the sample.

Documentation and batch traceability

With acylated analogues an analytical documentation should state, in addition to the batch number and method, the measured molecular mass, since the designation alone does not secure the structure. Notes on the structure of such documents can be found in the linked resources.

Storage and Stability

Lyophilised material is stored dry, protected from light and refrigerated. Amphiphilic peptide conjugates tend to aggregate in solution and to adsorb onto vessel surfaces, which can lead to under-recovery in quantitative measurements. These details concern exclusively the handling of laboratory substances.

Further research resources

LC-MS in peptide analysis

HPLC in peptide analysis

Understanding peptide purity

Analysis and certificates

Associated catalogue products

Semaglutide

Cagr 5mg + Sema 5mg

This page serves exclusively as scientific and analytical information.
