The bound metal determines colour, chromatographic behaviour and the requirements for documentation.

What is GHK-Cu?

GHK is the tripeptide glycyl-L-histidyl-L-lysine with a molecular mass of approximately 340 Da. GHK-Cu designates the complex of this tripeptide with divalent copper; the molecular mass of the complex is approximately 404 Da. Coordination takes place via the imidazole group of the histidine, the N-terminus and the amide bond.

The complex is characteristically blue in colour. This colour arises from d-d transitions of the copper(II) ion and is a simple but non-specific indication of the presence of complexed copper. Pure GHK without copper is colourless. Commercial material is present as a lyophilised blue powder.

Research context

GHK was originally described as a copper-binding peptide from human plasma and has since been the subject of cell biology and dermatology research literature. In cosmetic formulations GHK-Cu is established as an ingredient. For laboratory contexts the relevant question is above all in what ratio peptide and copper are actually present, since both under-complexed free GHK and excess copper salt can occur.

Analytical identification

For metal complexes the usual peptide analysis is not sufficient:

RP-HPLC is not unproblematic with GHK-Cu. Copper can interact with residual silanol groups and with the ion-pairing reagent, which causes peak tailing, retention drift or partial decomplexation on the column. Many laboratories therefore deliberately analyse the free peptide GHK under acidic conditions and determine the copper separately.

LC-MS confirms the peptide identity via the mass of the GHK moiety. Copper-containing species additionally show the characteristic isotope pattern of copper with the isotopes 63 and 65, which appears in the mass spectrum as a clearly recognisable double signal.

UV-Vis spectroscopy records the absorption band of the complex in the visible range and serves as a rapid indication of complex formation.

Elemental analysis such as ICP-MS or atomic absorption spectrometry determines the copper content quantitatively. This information cannot be derived from an HPLC chromatogram.

Purity and analytical interpretation

With GHK-Cu an area-percent value from HPLC describes only the organic fraction under the chosen conditions. It says nothing about whether the copper is present in stoichiometrically bound form. A complete characterisation of a copper complex therefore comprises at least the peptide purity, the peptide identity and the metal content. If only a single percentage value is given, it remains open what it refers to.

Documentation and batch traceability

For metal complexes an analytical documentation should state which method was used for the organic fraction and which for the metal, and assign both to the same batch. Without this separation a data sheet cannot be interpreted.

Storage and Stability

Copper complexes are sensitive to light and moisture. Common laboratory practice is to store the lyophilised material dry, protected from light and refrigerated. In aqueous solution copper can act as a redox catalyst and accelerate the oxidation of sensitive constituents; this concerns mixtures with other peptides in particular.

Further research resources

HPLC in peptide analysis

LC-MS in peptide analysis

How to read a Certificate of Analysis (COA)

Analysis and certificates

Associated catalogue products

GHK-Cu

AHK-Cu

GLOW Blend (GHK-Cu + TB-500 + BPC-157)

KLOW Blend (GHK-Cu + TB-500 + BPC-157 + KPV)

This page serves exclusively as scientific and analytical information.
