In fact it is the link between a physical container and all the data that exists about it. Without that link, every certificate of analysis is a document without a reference object.

Batch and lot: two terms, one principle

Both terms denote a defined quantity of material that was produced or combined under uniform conditions. The difference lies in the perspective.

Batch – the production unit: everything produced in one coherent manufacturing operation. Reference point for synthesis, purification and lyophilisation.

Lot – the dispensing or packaging unit: a defined sub-quantity that was filled or packaged together. Reference point for filling, packaging and shipping.

Combined identifier – linking of both levels, for example batch number plus fill identifier. Allows an individual container to be assigned to the production unit.

In practice, the terms are often used synonymously. What matters is not the designation, but whether the identifier is unambiguous and whether it appears identically on the container, on the certificate and in the internal documentation.

What is attached to a batch identifier

A batch number is a key. It points to a data record whose scope varies from manufacturer to manufacturer but typically comprises the following elements:

Date of manufacture or filling of the respective unit.

The associated analytical results for purity, identity and, where tested, content.

The test methods used, with their parameters, such as gradient and detection wavelength.

The certificate of analysis as a summarising document.

Details of packaging, container size and labelling.

In documented processes additionally the starting materials used, with their own batch identifiers.

The last point describes the chain upstream. Complete traceability links not only product and certificate, but also product and starting material. The extent to which this is documented differs considerably between suppliers.

The comparison: document against material

The practical check consists of holding several sources against one another. If they deviate from one another, the assignment is not secured, regardless of how good the figures in the certificate look.

Label against certificate – does the batch identifier on the container match the one in the document? A deviation means that the document may belong to a different unit.

Certificate against product designation – do the designation and identifier match the order? A deviation indicates a mix-up during picking or issuing.

Certificate date against date of manufacture – a certificate dated before the batch is not plausible.

Issuing body – a document without a named source is not verifiable.

Why a batch reference is what makes quality statements robust

A purity figure without a batch reference is a product description, not a measurement. Only the assignment to a specific unit turns it into a verifiable statement. That is the reason why batch-related documentation has a high status in quality management systems.

The second reason is the ability to delimit. If an anomaly is identified, the batch identifier makes it possible to determine which further containers could be affected. Without an identifier the entire stock would have to be considered.

Limits of traceability

A batch number documents assignment, not quality. It says nothing about how good a material is, only what the stated figures refer to.

Its informative value ends at the boundary of the documented process. What was not recorded cannot be reconstructed.

A certificate describes a sample at a point in time. Changes due to transport and storage are not included in it.

An identifier alone is not proof of authenticity. It only becomes verifiable once the issuing body can confirm the assignment.

Common misinterpretations

Treating a general sample document without a specific batch identifier as a batch-related certificate. That is a specification, not a COA.

Reading a batch number as a statement of quality.

Concluding from one tested batch to other batches of the same product.

Not recording the identifier at goods receipt, whereby the link to the storage location is lost.

Documentation context

This page considers the level before the certificate: the question of whether a document belongs to the material at hand at all. How the document itself is read section by section is described in Reading a COA correctly. The analytical methods behind the values are covered by HPLC in peptide analysis and LC-MS in peptide analysis; the systematics of the parameters by Understanding peptide purity. Batch-related documents are published under Analytics & Certificates insofar as corresponding real documents are available.

For laboratory research only.
